Quickly eyeballing the space where the spare tire is there should be plenty of volume for a 12" single. Rather than building a box you would want to mask off the bottom a fiberglass to the shape of the bottom. You could still build braces inside but the fiberglass molded will give you maximum volume. Just be sure to use a heavy duty grill

No spare tire - isn't that what roadside assistance is for?
